The Maxwell® CSC Rapid ccfDNA Kit enables fast and streamlined automated extraction of high-quality ccfDNA from 1–4ml of human plasma. Plasma is added directly into the prefilled Maxwell® CSC cartridges without any tedious manual sample pretreatment, significantly reducing hands-on time and minimizing effort. The preprogrammed Maxwell® CSC Rapid ccfDNA method with a run time of less than 30 minutes enables fully automated ccfDNA extraction while simultaneously processing 1 to 16 samples on the Maxwell® CSC Instrument or 1 to 48 samples on the Maxwell® CSC 48 Instrument, accelerating turnaround and boosting efficiency.

Prefilled Maxwell® CSC Rapid ccfDNA cartridges contain all the necessary reagents required for purification of ccfDNA by eliminating contaminants and inhibitors, delivering consistent results while minimizing hands-on steps and operator error. Compared to alternative ccfDNA extraction kits, ccfDNA extracted using the Maxwell® CSC Rapid ccfDNA Kit has a relatively lower level of genomic DNA carryover, reducing assay failures and enhancing confidence in your results. Streamlined Workflow with Minimal Hands-On Steps
The Maxwell® CSC Rapid ccfDNA Kit enables effortless ccfDNA extraction in three simple steps.
- Prepare Maxwell® cartridge: Simply load 1–4ml of untreated plasma into the sample binding wells, and add the supplied Proteinase K (PK2) Solution to well number 4 and a plunger to well number 6 of the new six-well Maxwell® CSC Rapid ccfDNA cartridge.
- Start automated extraction method: Place the prepared cartridges, complete with elution tubes containing 50µl of Elution Buffer (RCFD), in the Maxwell® CSC or Maxwell® CSC 48 Instrument, and launch the preprogrammed Maxwell® CSC Rapid ccfDNA method with touch of a button.

Flexible Workflow with Scalable Performance
The Maxwell® CSC Rapid ccfDNA Kit delivers concentrated ccfDNA with its recovery increasing proportionally across 1ml to 4ml plasma inputs. The linear scalability and flexibility to use a range of plasma input volumes in a single, streamlined workflow lets you work with varying input volumes available for different clinical samples without compromising ccfDNA yield, purity or downstream assay performance—your laboratory stays efficient and on-schedule even when sample volumes vary.
Linear increase in ccfDNA yield for different plasma input volumes. Plasma was isolated from whole blood collected in Vacutainer® K2EDTA tubes (K2EDTA plasma), and ccfDNA was extracted from 1–4ml of plasma input volumes using the Maxwell® CSC Rapid ccfDNA Kit. Purified ccfDNA was eluted in 50μl of Elution Buffer (RCFD), and exact eluate volume was measured. The ccfDNA yields were determined by a fluorometric assay using a dsDNA-specific dye and a qPCR assay for a 75bp DNA target. The ccfDNA yields scale linearly across 1–4ml of plasma input volumes. High-Quality and Pure ccfDNA for More Reliable Results and Maximized Success in Clinical Workflows
The Maxwell® CSC Rapid ccfDNA Kit isolates ccfDNA with relatively less genomic DNA carryover than other alternative kits. This offers a critical advantage when processing plasma drawn into standard blood collection tubes with K2EDTA anticoagulant or other non-stabilizing blood collection devices that can release leukocyte DNA. Cleaner ccfDNA inputs mean increased sensitivity and fewer failures in downstream assays.
High-yield and high-quality ccfDNA extracted from plasma collected in different blood collection tubes. Plasma was isolated from human whole blood collected in either Streck Cell-Free DNA BCT® (Streck plasma) or Vacutainer® K2EDTA tubes (K2EDTA plasma). ccfDNA was extracted from 4ml of plasma using either the Maxwell® CSC Rapid ccfDNA Kit or a manual competitor kit (Competitor Q) and eluted using 50µl of kit-specific Elution Buffer. ccfDNA yields were determined using either a fluorometric assay with a dsDNA-specific dye or a qPCR assay for amplification of a 75bp DNA target. Yields of ccfDNA extracted using the Maxwell® CSC Rapid ccfDNA Kit or the competitor kit are shown for Streck plasma (Panel A) and K2EDTA plasma (Panel B). Data represent mean ±SD, N=4. ccfDNA yields produced by the Maxwell® CSC Rapid ccfDNA Kit were comparable to or better than those from Competitor Q, except for slightly higher yields from K2EDTA plasma observed with Competitor Q in the qPCR assay, possibly due to contaminating genomic DNA. To assess ccfDNA quality, purified ccfDNA samples were analyzed by Agilent Cell-Free DNA ScreenTape Assay. Panel C. The TapeStation electropherogram for Competitor Q in Panel D shows a higher concentration of contaminating genomic DNA in ccfDNA extracted from K2EDTA plasma (indicated by red circle) as compared to the ccfDNA extracted using the Maxwell® CSC Rapid ccfDNA Kit. A large amount of genomic DNA carryover in ccfDNA extracted from K2EDTA plasma by Competitor Q resulted in higher reported yield of ccfDNA in the qPCR assay in Panel B, confounding the actual ccfDNA yield.

ccfDNA ready for digital PCR. ccfDNA samples purified from 4ml of plasma isolated from either Streck Cell-Free DNA BCT® (Streck Plasma) or Vacutainer® K2EDTA tubes (K2EDTA Plasma) using the Maxwell® CSC Rapid ccfDNA Kit were evaluated in a PIK3CA copy-number droplet digital PCR assay. Uniform 1-D droplet-amplitude plots across replicates of different individual samples demonstrate that ccfDNA eluates extracted using the Maxwell® CSC Rapid ccfDNA Kit are compatible with and deliver reproducible results in high-sensitivity dPCR workflows.
ccfDNA compatible with next-generation sequencing. ccfDNA was extracted from 4ml of plasma isolated from Streck Cell-Free DNA BCT® using either the Maxwell® CSC Rapid ccfDNA Kit or a competitor ccfDNA extraction kit (Competitor Q). Purified ccfDNA samples were analyzed in a Whole Genome Sequencing assay. Successful libraries were generated for all the samples, and sequencing was performed on a NovaSeq X Plus system (Illumina), using 2 × 150 paired-end sequencing in a shared-lane configuration, with 140–170M reads per library and subsampling of 70M reads per sample. A GC-bias plot was generated for ccfDNA extracted with the Maxwell® CSC Rapid ccfDNA Kit (yellow line) and Competitor Q (blue line) to determine how evenly AT- and GC-rich regions are covered using Picard CollectGcBiasMetrics tool (MultiQC: Summarized analysis results for multiple tools and samples in a single report. The curve for ccfDNA extracted using the Maxwell® CSC Rapid ccfDNA Kit tracks the ideal flat line (1.0) more closely at low and high GC, indicating lower bias and, therefore, more uniform coverage across the genome as compared to ccfDNA extracted with the competitor kit.
