The Maxwell® CSC XtractAll FFPE DNA/RNA Kit enables four distinct nucleic acid extraction workflows from FFPE tissues using a single, streamlined preprocessing protocol with just a few simple steps.

  1. Prepare FFPE Tissue Lysate: A simple pretreatment protocol streamlines FFPE tissue deparaffinization, lysis and nucleic acid decrosslinking in under 2 hours, reducing hands-on time and effort.
  2. Prepare Maxwell® Cartridge: Add FFPE tissue lysate and workflow-specific reagents as needed (refer to Technical Manual #TM755) and insert a plunger into Maxwell® CSC cartridge. Only one cartridge and plunger are needed per sample, regardless of the workflow, reducing plastic waste.
  3. Start automated extraction method: Load the prepared cartridges, with elution tubes containing 50µl of Nuclease-Free Water, in the Maxwell® CSC or Maxwell® CSC 48 Instrument, and start the workflow-specific preprogrammed Maxwell® CSC XtractAll FFPE method with the touch of a button.

Efficient recovery of high-yield nucleic acid from FFPE tissues across all workflows. DNA, RNA or Total Nucleic Acid were extracted from FFPE human lung tumor (Panel A) and FFPE human colon (Panel B) tissue samples using the four different workflows of the Maxwell® CSC XtractAll FFPE DNA/RNA Kit: 1) DNA, 2) RNA, 3) DNA/RNA sequential (‘Seq DNA’ and ‘Seq RNA’) and 4) total nucleic acid (TNA). DNA yield was determined using qPCR for amplification of a 102bp target, while RNA yield was evaluated by RT-qPCR amplifying a 100bp amplicon. Data shown as mean ± SD, N = 6. The TNA and DNA/RNA sequential workflows achieved comparable yields to singular DNA and RNA extractions, offering flexibility without compromising recovery.

High-yield and high-quality DNA and RNA extraction across diverse FFPE tissue types using the Maxwell® CSC XtractAll FFPE DNA/RNA Kit. DNA and RNA were extracted from a single section of each indicated FFPE human tissue type using either the Maxwell® CSC XtractAll FFPE DNA/RNA sequential workflow or two alternative manual kits following manufacturer protocols. DNA yield was assessed by qPCR targeting 150bp (Panel A) and 300bp (Panel B) amplicons. RNA yield was measured by RT-qPCR targeting 100bp (Panel C) and 171bp (Panel D) amplicons. Across all FFPE tissue types, the Maxwell® CSC XtractAll FFPE DNA/RNA Kit delivered comparable or higher DNA and RNA yields, particularly for longer amplicons, demonstrating its robust performance and effectiveness in recovering high-quality, amplifiable nucleic acids from challenging FFPE tissue samples. Superior DNA and RNA fragment quality from FFPE tissue using the Maxwell® CSC XtractAll FFPE DNA/RNA Kit. Detection of oncogenic variants by digital PCR using RNA extracted from FFPE tissue. RNA was extracted from different FFPE human lung tumor tissue samples using the Maxwell® CSC XtractAll FFPE DNA/RNA sequential workflow and analyzed using a droplet digital PCR assay for EML4-ALK mRNA fusions. DNA and RNA compatible with next-generation sequencing (NGS). DNA and RNA were extracted from a single section of FFPE human breast- and lung-tumor tissues using either the Maxwell® CSC XtractAll FFPE DNA/RNA sequential workflow (XtractAll) or a competitor FFPE DNA/RNA extraction kit (Competitor Q). Purified DNA and RNA samples were analyzed using the TruSight Oncology 500 (TSO 500) Assay (Illumina). Successful library preparation was achieved for all the samples, and sequencing was performed on the NovaSeq PE150 platform (Illumina), generating 80 million total reads per sample (40 million paired-end reads) at a coverage depth of 35,000X. Key quality control and NGS performance metrics for DNA and RNA extracted from the indicated FFPE tissue samples using the Maxwell® CSC XtractAll FFPE DNA/RNA Kit versus Competitor Q are presented. Compared to Competitor Q, the Maxwell® CSC XtractAll FFPE DNA/RNA Kit produced DNA and RNA with higher integrity and fragment length that exhibited better target coverage.

Reliable Fluorometric Quantitation that Translates to Downstream Assay Success

Fluorometric assays using DNA- and RNA-specific dyes are routinely used in clinical laboratories to estimate nucleic acid yield. Unlike other FFPE extraction methods, the Maxwell® CSC XtractAll FFPE DNA/RNA Kit delivers high-quality DNA and RNA whose fluorometric quantitation closely correlates with actual assay performance in downstream applications such as qPCR and digital PCR. Improved concordance between quantitation and amplification performance. Compared to alternative kits, the Maxwell® CSC XtractAll FFPE DNA/RNA Kit consistently demonstrated lower and narrower ratio distributions, indicating more reliable fluorometric quantitation and greater consistency in downstream amplification-based molecular assays.
